== Structure of penicillin binding protein 3 == Penicillin binding protein 3 is important for bacteria wall synthesis and is a main target in β-lactam antibiotics. It is a two-domain protein containing a C-terminal transpeptidase linked to an extended N-terminal domain. This protein is similar to other class B PBP’s since it contains an α-helical subdomain or “head” domain towards the N-terminus. The N-terminal domain’s function is still not known but it is thought it serves to position the transpeptidase domain away from the inner membrane as part of a multienzyme complex involved in cell wall biosynthesis.
These are commonly used in industry for the processing of metals such as the lanthanides; because the separation factors between the lanthanides are so small many extraction stages are needed. === Drug design of RXP 407 === RXP 407 is the first N-domain selective phosphinic peptide and was discovered by screening phosphinic peptides libraries. Before the discovery of RXP 407 it had long been claimed that the free C-terminal carboxylate group in P2’ position was essential to the potency of ACE inhibitor so it can be reasoned that this has postponed the discovery of N-domain selective ACE inhibitors. When RXP 407 was discovered researchers looked into phosphinic peptides with 3 different general formula, each containing 2 unidentified amino acids, only 1 of these general formula showed potent inhibition (Ac-Yaa-Pheψ(PO2-CH2)Ala-Yaa’-NH2). Peptide mixtures were made, substituting Yaa and Yaa’ with different amino acids, trying to establish if there would be a potent inhibitor that could inhibit either the N-domain or the C-domain of the enzyme. The result was that the compound Ac-Asp(L)-Pheψ(PO2-CH2)(L)Ala-Ala-NH2 actively inhibited the N-domain and was given the name RXP 407. Structure-function relationship showed that the C-terminus carboxamide group played a crucial role in the selectivity for the N-domain of ACE. Additionally, the N-acetyl group and the aspartic side chain in the P2 position aides in the N-domain selectivity of the inhibitor. These features make the inhibitor inaccessible to the C-domain but give good potency for the N-domain, this leads to a difference in inhibitory potency of the active sites of three orders of magnitude. These results also indicate that the N-domain possess a broader selectivity than the C-domain.

== Name and discovery == The name Coomassie was adopted at the end of the 19th century as a trade name by the Blackley-based dye manufacturer Levinstein Ltd, in marketing a range of acid wool dyes. In 1896 during the Fourth Anglo–Ashanti War, British forces had occupied the town of Coomassie (modern-day Kumasi in Ghana). In 1918 Levinstein Ltd became part of British Dyestuffs, which in 1926 became part of Imperial Chemical Industries. Although ICI still owns the Coomassie trademark, the company no longer manufactures the dyes. The blue disulfonated triphenylmethane dyes were first produced in 1913 by Max Weiler, who was based in Elberfeld, Germany. Various patents were subsequently taken out on the organic synthesis. Papers published in biochemistry journals frequently refer to these dyes simply as "Coomassie" without specifying which dye was used. The toxic action of mHTT may manifest and produce the HD pathology through multiple cellular changes. In its mutant (polyglutamine expanded) form, the protein is more prone to cleavage that creates shorter fragments containing the polyglutamine expansion. These protein fragments have a propensity to undergo misfolding and aggregation, yielding fibrillar aggregates in which non-native polyglutamine β-strands from multiple proteins are bonded together by hydrogen bonds. These aggregates share the same fundamental cross-beta amyloid architecture seen in other protein deposition diseases. The aggregates are covered on their surface with a 'fuzzy coat' that contains non-polyglutamine parts of the protein, as illustrated in a published structural model shown above. Over time, the aggregates accumulate to form inclusion bodies within cells, ultimately interfering with neuronal function. Inclusion bodies have been found in both the cell nucleus and cytoplasm.
